Description
English: Figure 1 Polatuzumab structure and mechanism of action. Polatuzumab is composed of an anti-CD79b monoclonal antibody conjugated to monomethyl auristatin E (MMAE) via a protease-cleavable linker. After binding to CD79b, the complex is internalized by endocytosis and directed to lysosomes where the linker is cleaved and MMAE released to the cytosol. Free MMAE inhibits microtubule formation and results in cell growth arrest and apoptosis. Extracellular MMAE release can occur leading to systemic adverse events.
